My current hypothesis is that the desire to orgasm (horniness) and the 'drop' afterward are both related to the level of 'fullness' of the prostate. Just as a full bladder results in the urge to go, a full prostate causes the urge to cum (play on words intended). A single orgasm/ejaculation does not empty the prostate, only a limited amount of fluid is released. It takes time to replenish the sperm/semen in the prostate, though the process is ongoing. By denying orgasm, the prostate fills more than it might otherwise, and a single orgasm/ejaculation may not reduce the volume below (or not much below) the level that we perceive as being horny, and no 'drop' occurs (or is lessened), despite the release of the chemical signals that would normally result in a 'drop'. The level of fullness overrides the chemical signal.
Correspondingly, two or more orgasms/ejaculations that deplete the level in a short span of time releases the physical override and lets the chemical signals do their job, which is to reduce the desire for sex/orgasm until the level of semen returns to optimum.
(This might be an interesting line of research for a Doctoral thesis, if one were able to devise a method of reliably quantifying 'desire' such that it was not subject to the bias of an individual's perception of it.)
